Diamine oxidase (DAO, EC1.4.3.6) is widely present in animals (intestinal mucosa, lungs, liver, kidneys, etc.), plants and microorganisms. It can catalyze the oxidation of polyamines to aldehydes. Its activity is closely related to the synthesis of nucleic acids and proteins, and can reflect the integrity and degree of damage of the intestinal mechanical barrier.
DAO can catalyze cadaverine to produce aldehyde and hydrogen peroxide. After adding excess horseradish peroxidase, it can catalyze hydrogen peroxide to oxidize o-benzidine to produce a colored substance. The colored substance has characteristic absorption at 500nm. The activity of DAO can be calculated by measuring the increase rate of absorbance at this wavelength.
